چکیده

Metabolic actions of insulin are mediated by the insulin receptor/insulin receptor substrate (IRS)-1/ phosphatidylinositol 3-kinase signaling pathway in insulin target organs including the liver, skeletal muscle and adipose tissue. To understand the actual pathway of IRS-1 is the scientists’ hope for better understanding the disease mechanism. In this work, a systematic network creation for human insulin receptor substrate 1 by systemic biology technique was performed. According to this work, there are at least 9 genes directly related to IRS-1 gene. Of interest, the relation of diabetes to some identified genes can be seen. In addition, identified relation to some genes implies a correlation between IRS-1 and cancer. The systematic network of IRS-1 created in this study could provide useful data for further works in diabetic and oncologic medicine. INTRODUCTION

Accelerated atherosclerosis in Apoe-/- mice heterozygous for the insulin receptor and the insulin receptor substrate-1.

OBJECTIVE Prediabetic states are associated with accelerated atherosclerosis, but the availability of mouse models to study connections between these diseases has been limited. The aim of this study was to test the selective role of impaired insulin receptor/insulin receptor substrate-1 signaling on atherogenesis. METHODS AND RESULTS To address the effects of impaired insulin signaling associ...

The Drosophila insulin receptor activates multiple signaling pathways but requires insulin receptor substrate proteins for DNA synthesis.

The Drosophila insulin receptor (DIR) contains a 368-amino-acid COOH-terminal extension that contains several tyrosine phosphorylation sites in YXXM motifs. This extension is absent from the human insulin receptor but resembles a region in insulin receptor substrate (IRS) proteins which binds to the phosphatidylinositol (PI) 3-kinase and mediates mitogenesis. The function of a chimeric DIR cont...

Defects of the insulin receptor substrate (IRS) system in human metabolic disorders.

Insulin receptor substrate (IRS) molecules are key mediators in insulin signaling and play a central role in maintaining basic cellular functions such as growth, survival, and metabolism. They act as docking proteins between the insulin receptor and a complex network of intracellular signaling molecules containing Src homology 2 (SH2) domains. Four members (IRS-1, IRS-2, IRS-3, IRS-4) of this f...

The Importance of α-CT and Salt bridges in the Formation of Insulin and its Receptor Complex by Computational Simulation

Insulin hormone is an important part of the endocrine system. It contains two polypeptide chains and plays a pivotal role in regulating carbohydrate metabolism. Insulin receptors (IR) located on cell surface interacts with insulin to control the intake of glucose. Although several studies have tried to clarify the interaction between insulin and its receptor, the mechanism of this interaction r...
